Dunno about being the first crossdressing movie, but it could certainly be the first Benny Hill chase!

Men in women’s clothing were something of a staple of English music hall and translated very quickly onto film, where the possibilities for mayhem gave delighted audiences a comic frisson.
It may well have been one of the first, if not the first.
